Expectation and Dopamine

چکیده
The power of placebos has long been recognized for improving numerous medical conditions such as Parkinson’s disease (PD). Little is known, however, about the mechanism underlying the placebo effect. Using the ability of endogenous dopamine to compete for [C]raclopride binding as measured by positron emission tomography, we provide in vivo evidence for substantial release of endogenous dopamine in the striatum of PD patients in response to placebo. Our findings indicate that the placebo effect in PD is powerful and is mediated through activation of the damaged nigrostriatal dopamine system.

Investigating expectation and reward in human opioid addiction with [11C]raclopride PET
The rewarding properties of some abused drugs are thought to reside in their ability to increase striatal dopamine levels. Similar increases have been shown in response to expectation of a positive drug effect. The actions of opioid drugs on striatal dopamine release are less well characterized. We examined whether heroin and the expectation of heroin reward increases striatal dopamine levels i...
متن کاملAt-risk for pathological gambling: imaging neural reward processing under chronic dopamine agonists.
Treatment with dopamine receptor agonists has been associated with impulse control disorders and pathological gambling (PG) secondary to medication in previously unaffected patients with Parkinson's disease or restless legs syndrome (RLS). In a within-subjects design, we investigated the underlying neurobiology in RLS patients using functional magnetic resonance imaging. We scanned 12 female RL...
متن کاملEffects of expectation on placebo-induced dopamine release in Parkinson disease.
CONTEXT Expectations play a central role in the mechanism of the placebo effect. In Parkinson disease (PD), the placebo effect is associated with release of endogenous dopamine in both nigrostriatal and mesoaccumbens projections, yet the factors that control this dopamine release are undetermined. OBJECTIVE To determine how the strength of expectation of clinical improvement influences the de...
متن کاملEver-Changing Cycles of Musical Pleasure: The Role of Dopamine and Anticipation
Music listening is highly pleasurable and important part of most people’s lives. Because music has no obvious importance for survival, the ubiquity of music remains puzzling and the brain processes underlying this attraction to music are not well understood. Like other rewards (such as food, sex, and money), pleasurable music activates structures in the dopaminergic reward system, but how music...
متن کاملDynamic mesolimbic dopamine signaling during action sequence learning and expectation violation
Prolonged mesolimbic dopamine concentration changes have been detected during spatial navigation, but little is known about the conditions that engender this signaling profile or how it develops with learning. To address this, we monitored dopamine concentration changes in the nucleus accumbens core of rats throughout acquisition and performance of an instrumental action sequence task. Prolonge...
متن کاملEvaluation of iterative reconstruction method and attenuation correction on brain dopamine transporter SPECT using anthropomorphic striatal phantom
Objective(s): The aim of this study was to determine the optimal reconstruction parameters for iterative reconstruction in different devices and collimators for dopamine transporter (DaT) single-photon emission computed tomography (SPECT). The results were compared between filtered back projection (FBP) and different attenuation correction (AC) methods. Methods: An anthropomorphic striatal phant...
